Winger Arjen Robben will definitely miss the Netherlands' opening World Cup match because of the torn hamstring sustained against Hungary on Saturday.
Dutch boss Bert van Marwijk said the 26-year-old was having intensive physio but would not face Denmark on Monday.
"Whatever one may say, his injury is serious and I do not know when he can join us," said Van Marwijk.
"The team's goal now is to go as far as possible to allow Arjen to play with us during this World Cup."
Former Chelsea and Real Madrid winger Robben sustained what is described as a "small tear" in his left hamstring towards the end of Saturday's 6-1 warm-up victory over Hungary.
The second-half substitute had scored two goals before pulling up when in possession in the 85th minute of his country's final warm-up match in Amsterdam.
On Monday, Robben, who was voted Germany's Footballer of the Year after excelling for Double winners Bayern, said he was "very hopeful" he would recover in time to play some part in the World Cup.
"It is impossible to say whether [a return] will only be about four days or a week away, but we will do everything to get fit. I'm very hopeful," he added.
The Netherlands begin their Group E campaign against the Danes in Johannesburg, followed by games against Japan on 19 June and Cameroon on 24 June.

Benfica midfielder Ruben Amorim will replace Nani in Portugal's World Cup squad, the Portuguese Football Federation (FPF) has confirmed.

The Manchester United winger had missed Saturday's training session by precaution after sustaining a shoulder injury which did not look serious enough to rule him out of the tournament.

However, medical tests undergone this morning to learn the real extent of the problem revealed Nani will not be able to take part in Portugal's World Cup campaign.

An FPF statement read: "Following the injury involving the left clavicle sustained by the player Nani and after monitoring his progress on a daily basis, with the completion of additional tests, we have concluded the competitive inability of the player. The medical file will be presented to the medical department of Manchester United."

The decision comes as a major blow to Portugal's chances in South Africa as Nani, who scored against Cameroon a week ago, had been in scintillating form of late.

Amorim had been on stand-by since Friday, and will board a plane to South Africa still today to join up with the rest of the Seleccao.

The 25-year-old centre midfielder, who can also operate on the right side of the defence and midfield, has been called up for the first time to represent Portugal's main squad, having previously played for the Seleccao's Under-21 and B squads.

Portugal, who find themselves in Group G alongside Brazil, North Korea and Cote d'Ivoire, will kick off their campaign against the Elephants on June 15th.

France midfielder Lassana Diarra has been ruled out of this summer's World Cup with a stomach injury.
The Real Madrid star, 25, has been ordered to rest for an "indefinite period", according to a statement from the French Football Federation.
Diarra, who made coach Raymond Domenech's 24-man shortlist for his 23-strong World Cup squad, suffered the setback on a team trek in Tignes.
A replacement has not yet been called up for the former Portsmouth player.
"Medical tests have revealed an unforeseeable illness which justifies a rest period," said France team spokesman Francois Manardo.
"As a consequence, Lassana Diarra will miss the World Cup."
Domenech has until 1 June to submit his final selection to tournament organisers Fifa, but can include players from outside the group of 24 he named on Monday.
"It's a blow when you lose a player. It's even harder because it wasn't a choice, it's destiny," said Domenech.
"It's tough for him because I know how much effort he had put into this World Cup. He was one of the driving forces of this team in terms of the way he played and his attitude."
Diarra's injury might force a recall as he is one of only seven midfielders in Domenech provisional group.
The anchorman played in 10 of France's 12 matches in their 2010 World Cup qualification campaign.
Diarra, who also played for Arsenal and Chelsea during his time in England, missed training on Friday and Saturday after picking up the injury whilst altitude training on a mountain glacier on Thursday.
Although France open their campaign against Uruguay at sea level in Cape Town on 11 June, they then travel over 1000m up to play Mexico and South Africa in Polokwane and Bloemfontein respectively.
Since his injury Diarra's defensive role has been filled by Arsenal anchorman Abou Diaby in training.
Domenech is unlikely to risk a repeat of Euro 2008, when he selected Patrick Vieira even though the midfielder had not fully recovered from a thigh injury.
Vieira never recovered and did not play in the tournament, where France went out in the group stages without winning a game.
